Test compounds (50-0.2 micro molar) or postive control (5.0.02 micro molar), cofactors mix, and G-6-PDH were added to the wells and pre incubated for 10 min at 37 degrees celsius, and the plates were read to account for any fluorescence caused by the test compounds (0-min readouts, both diosgenin and dioscin had no measurable auto fluorescence). Reaction was initiated by adding the enzyme substrate mixtre and incubated for 30 min. Reaction was stopped by adding ice cold acetonitrile/0.5M Trisbase (80:20).

GM and ZO exhibitied moderate potencies (mean IC50 11.33 and 11.58 microgram/mL, respectively).
