Remark : The ex vivo results showed that the formation rates of 6-beta-hydroxytestosterone from testosterone significantly decreased in a dose-dependent manner after pretreatment of rats with PWT for 7 d compared with the control group. Formation rates decrease gradually with the increasing doses of PWT. CYP3A protein and mRNA levels were also markedly down-regulated by PWT treatment. No marked effect or only a slight inhibition of CYP3A activity ex vivo was observed after the same treatment of WTD, which significantly reversed the inhibition caused by PWT single herb. CYP3A protein and mRNA levels were significantly higher in WTD groups than in PWT groups at the tested doses.
Note : PWT = processed Wutou (Aconitum carmichaelii) WTD = Wutou decoction consists of five medicinal herbs, Radix Aconiti, Herba Ephedrae, Radix PaeoniaeAlba, Radix Astragali, and Radix Glycyrrhizae.
