Remark : Result: Neferine showed a significant mechanism-based inhibition of CYP3A4. Neferine displayed Pi bonds with the heme group of CYP3A4, which may produce a mechanism-based inhibition by the heme group pathway. Combined with the CYP3A4 substrate, it is reasonable to believe that Nef has a mixed-type inhibition or positive cooperativity of CYP3A4 mediated testosterone metabolism.
Note : Phenacetin, dextromethorphan and testosterone, respectively, were used as CYP1A2, CYP2D6 and CYP3A4 substrates, and their metabolites were determined by validated HPLC methodologies.
